EpCAM, a carcinoma cell-surface marker protein and a therapeutic target, has been primarily addressed as a cell adhesion molecule. With regard to recent discoveries of its role in signaling with implications in cell proliferation and differentiation, and findings contradicting a direct role in mediating adhesion contacts, we provide a comprehensive and updated overview on the available structural data on EpCAM and interpret it in the light of recent reports on its function. First, we describe the structure of extracellular part of EpCAM, both as a subunit and part of a cis-dimer which, according to several experimental observations, represents a biologically relevant oligomeric state. Next, we provide a thorough evaluation of reports on EpCAM as a homophilic cell adhesion molecule with a structure-based explanation why direct EpCAM participation in cell-cell contacts is highly unlikely. Finally, we review the signaling aspect of EpCAM with focus on accessibility of signaling-associated cleavage sites.

In the present study, to evaluate neurobehavioral toxicity and the thyroid-disrupting effects of environmental levels of triphenyltin (TPT), the zebrafish larvae were exposed to 1, 10 and 100 ng/l TPT. In the neurobehavioral assay, increased levels of dopamine and serotonin, decreased content of nitric oxide, inhibited activities of acetylcholinesterase and monoamine oxidase were observed in the whole body of zebrafish larvae after TPT treatment, as well as the serious abnormal non-reproductive behavior. Moreover, the whole-body the T4 levels were markedly decreased significantly, whereas T3 levels were not significantly changed under TPT stress. In addition, TPT exposure significantly changed the expression levels of genes related to thyroid system, including corticotropin-releasing hormone gene crh, thyroid-stimulating hormone gene tsh beta, thyroglobulin gene tg, sodium/iodide symporter gene nis, thyroid hormone nuclear receptor Era, isoform tr beta, types I deiodinase gene dio1and types Il deiodinase gene dio2. The regulated responsiveness of thyroid hormone and related genes expression levels suggested that TPT could induce the thyroid disrupting effects in zebrafish larvae. Therefore, our results provide new aspects of TPT as an endocrine disrupting chemical.
