Recombinant viruses are especially useful for gene transfer and protein expression. Knowing the titre of a recombinant virus stock is really important prior to testing for protein production in insect cells. Each new lot of protein and each new set of experimental conditions should be tested in a titration (dose response) experiment. This needs to be done when a new experiment is set up or with a new lot or source of protein. Traditionally, the titre of a recombinant protein is routinely measured using cell proliferation assay, enzyme assay, or a plaque-forming unit assay (PFU). These methods are time-consuming, require a long infection period, and suffer from a high degree of inter-assay variability and are affected by virus-cell interactions.
